We offer ELISA assays in multiple formats, including direct, indirect, standard sandwich, and competitive ELISA, as well as ELISpot and In-cell ELISA.
The ELISpot assay is more sensitive than traditional ELISA or intracellular cytokine staining. Its high sensitivity comes from plate-bound antibodies that directly capture secreted proteins before they are diluted, degraded, or interact with cell-surface receptors. This enables the detection of very low frequencies of cytokine-secreting cells and supports high-throughput screening.
Spots are detected using a precipitating substrate, producing visible footprints of individual protein-secreting cells on the membrane. The method is highly effective and stable, with spots retaining integrity for years, allowing ELISpot plates to be reanalyzed even after long-term storage at room temperature.
ELISpot is especially valuable for assessing cell-mediated immunity, offering exceptional sensitivity and specificity for rare antigen-specific cells. It provides quantitative insights into cytokine secretion and other immune mediators at the single-cell level.
ELISpot is widely applied in immunogenicity testing, immune monitoring, epitope discovery, and T cell response evaluation. It works with a variety of cell suspensions, including PBMCs, lymphoid tissue, spleen, bone marrow, and CNS samples.
For multiplex applications, FluoroSpot—an advanced version of ELISpot—enables simultaneous detection of multiple cytokines secreted by a single T cell using fluorophore-labeled antibodies. This is especially useful for profiling T cell subpopulations with diverse cytokine patterns.
We also employ RAWspot™ technology with IRIS, ensuring precise detection of every spot. This system minimizes variability, integrates 3D spot volume (RSV) analysis for accurate quantification, and maintains reliability even in crowded wells
